چکیده
The super healer Murphy Roths Large (MRL) mouse represents the “holy grail” of mammalian regenerative model to decipher key mechanisms that underlies regeneration in mammals. At a time when mesenchymal stem cell (MSC)-based therapy most promising approach treat degenerative diseases such as osteoarthritis (OA), identification factors responsible for potential MSC derived from MRL would be major step forward medicine. In present study, we assessed and compared (MRL MSC) C57BL/6 (BL6 mice. First, compare phenotype differentiation BL6 did not observe any difference. Then, evaluated proliferation migration cells found while proliferate at slower rate than MSC, they migrate significantly higher rate. This is mediated, part, by MSC-secreted products since conditioned medium contains complex released increased MSC. A comparative analysis secretome quantitative shotgun proteomics Western blotting revealed produce release levels mesencephalic astrocyte-derived neurotrophic factor (MANF) BL6, BALB/c, DBA1 MANF knockdown using specific small interfering RNA (siRNA) reduced both scratch wound assay their ear punch Finally, injection silenced protect mice OA development. conclusion, our results evidence enhanced protection might be, attributed an effective reservoir MANF.

Regenerative Effect of Bone Marrow-derived Mesenchymal Stem Cells in Thioacetamide-induced Liver Fibrosis of Rats
The present study determined the regenerative effect of bone marrow-derived stem cells (BMSCs) on thioacetamide (TA)-induced liver fibrosis in rats. A total of 30 male Wistar rats were randomly divided into sham control and treatment groups. The rats of the sham control group were subdivided into three groups and sampled on the 14th, 18th, and 20th weeks after fibrosis induction. The rats of th...
متن کاملEffects of BIO on proliferation and chondrogenic differentiation of mouse marrow-derived mesenchymal stem cells
In vitro expansion of mesenchymal stem cell (MSCs) into large number is necessary for their application in cell-based treatment of articular cartilage defects. On the other hand, some studies have indicated that BIO (6-Bromoindirubin-3-Oxime) possesses mitogenic effects on cell culture. The objective of the present study was to examine the effect of BIO on in vitro expansion and chondrogenic di...

In the recent years, the unique characteristics of mesenchymal stem cells (MSCs), consisting their angiogenesis and vascularization activity, as well as anti-inflammatory, anti-ulcer and immunosuppressive features plusdifferentiation capability into multilineage cell, have provided huge favorite among clinicians and researchers that theirs experiments has focused on treatment of different disea...
متن کاملCholinergic Differentiation of neural precursor cells derived from mouse embryonic stem cells increased by Shh, LIF and RA
Introduction Cholinergic system is one of the important systems of mammalian CNS. Cholinergic neurons distributed in brain and spinal cord and contributed to principal functions like: consciousness, learning and memory, and motor control. In this study we investigated the differentiation potentiality of mouse embryonic stem cells toward cholinergic neurons. The aim of this study was to evaluate...
متن کاملHuman Mesenchymal Stem Cells Derived from Adiopose Tissue and Placenta and the Adipocytic and Osteocytic Differentiation
Introduction: Mesenchymal stem cells can be isolated from adult tissues, such as the adipose tissue, or other sources. Among all these sources, adipose tissue because of easy access, and placenta due to its immunomodulatory properties, in addition to another useful properties, were attracted more attention to themselves. Isolation and comparing these two different sources can help us for acces...
